UJI FENOLIK TOTAL DAN KAPASITAS ANTIOKSIDAN METODE ABTS DAUN KEDONDONG (SPONDIAS DULCIS PARKINSON)

Kedondong (Spondias dulcis Parkinson) merupakan tanaman tropis yang tersebar luas di Asia Tenggara dan telah lama dimanfaatkan secara tradisional untuk mengatasi infeksi kulit, batuk, demam, serta nyeri tenggorokan, juga sering dikonsumsi sebagai sayuran. Daunnya diketahui mengandung berbagai senyawa bioaktif seperti flavonoid, tanin, fenolik, saponin, triterpenoid, dan vitamin C yang berperan sebagai antioksidan alami dalam menetralisir radikal bebas, termasuk ROS dan RNS, pemicu stres oksidatif dan kerusakan seluler. Penelitian ini menganalisis total senyawa fenolik dan aktivitas antioksidan dari ekstrak metanol daun kedondong secara in vitro. Penentuan kandungan fenolik total dilakukan dengan metode Folin-Ciocalteu, sedangkan evaluasi kapasitas antioksidan menggunakan uji ABTS. Hasil penelitian menunjukkan bahwa ekstrak metanol daun kedondong memiliki kandungan fenolik total sebesar 54.691 mg GAE/g serta potensi aktivitas antioksidan tinggi dengan nilai IC₅₀ sebesar 16.05 µg/mL. Menurut klasifikasi aktivitas antioksidan oleh Ranggaini et al. (2024), antioksidan dikategorikan sangat kuat jika nilai IC₅₀ < 50 µg/mL, kuat 50–100 µg/mL, sedang 101–150 µg/mL, dan lemah >150 µg/mL. Dengan nilai IC₅₀ sebesar 16.05 µg/mL dan kandungan fenolik yang tinggi, ekstrak metanol daun kedondong menunjukkan aktivitas antioksidan sangat kuat serta berpotensi dikembangkan sebagai sumber antioksidan alami untuk perlindungan terhadap kerusakan akibat radikal bebas.References
