PENGARUH PEMBERIAN PROBIOTIK TERHADAP GAMBARAN HISTOPATOLOGI HEPAR PADA TIKUS DIET TINGGI LEMAK

Pemberian makanan tinggi lemak pada tikus dapat secara spontan mendorong terjadinya NASH, fibrosis, dan NAFLD yang menginduksi perubahan mikrobiota usus dan metabolit pada tikus. Penggunaan bahan pangan fungsional suplemen probiotik merupakan alternatif terapi komplementer, dengan kandungan senyawa bakteri asam laktat (BAL) yang mampu menurunkan kadar kolesterol. Studi menggunakan desain penelitian Posttest Only Group Design dengan 15 sampel Bahan Biologik Tersimpan (BBT) hepar tikus galur wistar jantan dari 5 kelompok. Kelompok kontrol negatif dengan pakan standar, kelompok kontrol positif dengan diet tinggi lemak, kelompok simvastatin dengan diet tinggi lemak, kelompok probiotik dengan diet tinggi lemak, dan kelompok kombinasi simvastatin dan probiotik dengan diet tinggi lemak. Dengan melakukan pengamatan secara histopatologi untuk melihat dan membandingkan efek probiotik dan simvastatin terhadap struktur seluler dan jaringan hepar. Pada kelompok kontrol negatif secara keseluruhan ditemukan hepatosit normal dengan sedikit ditemukan degenerasi balon dan tidak terdapat inflamasi lobular. Kelompok kontrol positif secara keseluruhan ditemukan steatosis dengan total kurang dari 5% dengan sedikit degenerasi balon dan beberapa fokus inflamasi lobular. Kelompok simvastatin seluruh sampel mengalami sedikit steatosis dengan variasi jumlah fokus inflamasi lobular dan degenerasi balon. Kelompok probiotik menunjukkan hasil yang lebih bervariasi dan secara umum lebih baik dibandingkan kontrol positif. Kelompok kombinasi simvastatin dan probiotik menunjukkan hasil terbaik dengan tingkat steatosis menurun secara signifikan dengan sedikit degenerasi balon dan satu sampel yang terdapat inflamasi lobular dengan score 2 menurut NASH CRN. Kesimpulan dari penelitian diperoleh, hasil pemberian secara kombinasi suplementasi probiotik dan simvastatin mampu menurunkan jumlah perlemakan, inflamasi lobular, dan degenerasi balon pada tikus percobaan.References
